6x-His hG-CSF with TEV Cut Site & PpiB Solubility Factor

This composite part consists of a T7 promoter with RBS (BBa_K320005), followed by a 6x N-Terminus His Tag (BBa_K320003), followed by a coding region for the solubility enhancer PpiB (BBa_K320000), a TEV cleavage site (BBa_K320002), and hG-CSF (BBa_K320001), followed by a stop codon, spacer, and T7 terminator (BBa_K320006).

This construct produces a fusion protein with an N-Terminus his tag, the solubility enhancer PpiB, a TEV cleavage site, and the protein hG-CSF.

The his tag allows for his tag purification, the PpiB increases solubility, TEV is site that allows you to cleave the hG-CSF from the PpiB and his tag using a protease, and hG-CSF is a drug used to stimulate white blood cell growth.

Because of the T7 promoter and RBS, for protein production this construct must be used in T7 cell lines like BL21 that produce T7 RNA polymerase.
